摘要:预处理是竹材酶水解的关键步骤,绿液预处理是一种通过改变木质纤维素表面物理和化学性质来提高酶水解糖化效率的有前景的方法,目前通过两段预处理定量分析竹材木质纤维素酶解糖化率的工作却很少。文章中主要采用绿液、热水以及热水+绿液等预处理方法,结合BSA、Tween80等化学助剂对粉单竹进行处理,使用纤维素酶对处理后的样品进行酶水解,探究预处理以及化学助剂对粉单竹酶水解效率的影响因素。结果表明,热水+绿液两段预处理对竹材的酶水解作用非常明显,HW1+GL4预处理后可使酶水解总糖得率达到85.51%,添加助剂BSA和Tween80可以对提高竹材的酶解糖化效率非常明显,两者协同作用可使酶水解总糖得率达到99.67%。 |
Abstract:Pretreatment is a key step in the enzymatic hydrolysis of bamboo, and green liquor pretreatment is a promising method to improve the enzymatic hydrolysis saccharification efficiency by changing the physical and chemical properties of lignocellulose surface. In this paper, the pretreatment methods such as green liquor, hot water and hot water + green liquor were mainly used, combined with chemical additives such as BSA and Tween80, and the enzymatic hydrolysis of the treated samples was carried out by cellulase, so as to explore the influencing factors of pretreatment and chemical additives on the enzyme hydrolysis efficiency of bamboo. The results showed that the enzymatic hydrolysis effect of hot water + green liquor on bamboo was very obvious, and the yield of total enzymatic hydrolysis of total sugar could reach 85.51% after HW1+GL4 pretreatment, and the addition of additives BSA and Tween80 could improve the enzymatic hydrolysis and saccharification efficiency of bamboo, and the synergistic effect of the two could make the enzymatic hydrolysis of total sugar yield reach 99.67%. |
